Revisiting Sertoli Cell Gene Expression and Dioxin Exposure: Cytochrome P450, AhR Signaling, and Molecular Mechanisms of Reproductive Toxicity — A Retrospective Critical Review (2004–2026) Copy title
By Judy Staveley.
Abstract
This retrospective critical review revisits the scientific questions underlying a 2003–2004 grant-supported investigation, “Looking at Gene Expression in Sertoli Cells,” conducted by Dr. Judy Staveley at the University of Alaska Anchorage. Drawing on foundational and contemporary scientific literature, the manuscript examines dioxin-associated molecular responses in the testis, with particular emphasis on aryl hydrocarbon receptor (AhR) signaling, cytochrome P450 regulation involving CYP1A1 and CYP1B1, oxidative stress, endocrine responsiveness, blood–testis barrier integrity, Sertoli-cell immunoregulation, and reproductive toxicology. The review explores advances in molecular biology and transcriptomic technologies from 2004 through October 2026, while critically evaluating the limitations of existing evidence across experimental models, exposure conditions, developmental stages, and cellular populations. Particular attention is given to distinguishing changes in gene expression from genetic mutations and separating mechanistic plausibility from experimentally demonstrated reproductive dysfunction. The original laboratory report and primary experimental data from the 2003–2004 investigation are not currently available. Accordingly, this manuscript does not reconstruct historical experimental findings or present new laboratory or computational analyses. This independent scholarly review identifies continuing research questions and future directions for investigating environmental toxicants, Sertoli-cell homeostasis, and male reproductive health.
