Perspectives and Insights on Neuroactive Steroid Antidepressant Drugs: From Development to Submission of New Drug Submission (NDS) in Canada
By Meera Chauhan, Maitry Sur and Peivand Pirouzi.
Abstract
Neuroactive steroid antidepressants are a novel class of central nervous system agents, first exemplified by an intravenous formulation and later advanced by an orally administered agent, the first medication in this class specifically authorized for postpartum depression (PPD) in Canada. PPD affects up to one in five Canadian women and is a leading cause of maternal mortality, making effective, rapidly acting treatment a critical unmet need. Unlike conventional serotonergic antidepressants, these agents act on the brain's inhibitory GABA system and produce symptom relief within days rather than weeks. This article examines the class's clinical use, chemical properties, packaging, safety profile, emerging innovations, and the regulatory pathway for preparing a New Drug Submission (NDS) to Health Canada, illustrating how mechanistic novelty shapes regulatory strategy for maternal mental health therapeutics.
