Abstract
Purpose
Acute promyelocytic leukemia (APL) is effectively treated with arsenic trioxide (ATO) and all-trans retinoic acid (ATRA), but arsenic-associated systemic toxicity remains an important clinical concern. This study aimed to develop a traditional Chinese medicine-inspired arsenic-doped carbon dot nanoplatform for APL therapy.
Methods
Arsenic-doped carbon dots (As-CDs) were prepared by thermal pyrolysis using Jujubae fructus as the biomass carbon precursor and Arsenic sublimate as the arsenic source. Their physicochemical properties, arsenic speciation, cellular uptake, cytotoxicity, reactive oxygen species (ROS) generation, mitochondrial membrane potential, apoptosis, differentiation-associated effects, pharmacokinetics, in vivo anti-leukemic efficacy, and biosafety were systematically evaluated.
