Expression of TTP in esophageal adenocarcinoma and its inhibition on epithelial-mesenchymal transition in esophageal adenocarcinoma cells
By Fang Z, Chen X and Luo Q.
Abstract
Objectives
Tristetraprolin (TTP), an important RNA-binding protein, regulates cell proliferation and differentiation by degrading target mRNAs; however, its role in the development and progression of esophageal adenocarcinoma (EAC) remains unclear. This study aims to detect the expression of TTP in EAC tissues and its relationship with clinicopathological features, and to investigate the effects of TTP overexpression on the proliferation of EAC cells and its underlying molecular mechanisms, thereby providing a basis for elucidating the role of TTP in the development and progression of EAC.
Methods
Twenty paraffin-embedded EAC specimens and their matched adjacent normal tissues were collected from the Department of Pathology, Chongqing University Cancer Hospital. TTP expression was compared between tumor and paired normal tissues by immunohistochemistry. The association between TTP expression and clinicopathological parameters (age, sex, tumor differentiation, smoking, alcohol consumption, pathological stage, and lymph node metastasis) was analyzed. Esophageal adenocarcinoma OE33 cells were infected with a recombinant lentivirus carrying a tet-on-TTP construct, and TTP overexpression following doxycycline (Dox) induction was verified by Western blotting and real-time PCR. The effect of TTP on EAC cell proliferation was assessed using the CCK-8 assay. Expression of epithelial-mesenchymal transition (EMT)-related proteins (E-cadherin, N-cadherin, Vimentin, and Snail) was determined by Western blotting.
