Thermodynamic Allosteric Stabilization and Oral Bio-Encapsulation for 1q21 GBA1 Gaucher Disease: Overcoming the \Delta\Delta G_{fold} Unfolding Barrier, Eliminating the Lyso-GL1 Myeloma Clonal Hazard, and Hardware-Gating Statutory Subsidy Continuity via PHOENIX_ROUTER

Abstract
Standard-of-care intravenous Enzyme Replacement Therapy (ERT; imiglucerase, velaglucerase alfa) for Gaucher disease (chromosome 1q21 GBA1 deficiency; OMIM 230800, 230900, 231000) imposes an unsustainable macroeconomic and household burden—exceeding JPY 160,000 per month in out-of-pocket patient co-payments upon loss of pediatric coverage and JPY 25,000,000–45,000,000 annually in gross national health expenditure per patient. This cost structure stems from three fundamental bottlenecks: (1) mammalian Chinese Hamster Ovary (CHO) bioreactor cultivation coupled with multi-step exoglycosidase glycan remodeling to expose terminal alpha-D-mannose residues; (2) severe thermodynamic destabilization (\Delta\Delta G_{fold} = +4.8 \pm 0.4 kcal/mol) in East Asian prevalent neuropathic/visceral alleles such as p.Leu483Pro (L444P) and RecNciI, which drives premature endoplasmic reticulum-associated degradation (ERAD) and rapid plasma inactivation; and (3) catastrophic splenic and bone-marrow accumulation of deacylated glucosylsphingosine (Lyso-GL1) via acid ceramidase (ASAH1) whenever treatment access u_{tx}(a) is interrupted at the age-18/20 statutory pediatric-to-adult transition cliff, exponentially accelerating CD1d-mediated Type II NKT cell stimulation, monoclonal gammopathy of undetermined significance (MGUS), and multiple myeloma (MM) somatic clonal evolution h_{som}(a). Building directly upon the thermodynamic foundations of Treatise V, this monograph establishes Treatise VI: a rigorous mathematical, pharmacokinetic/pharmacodynamic (PK/PD), and hardware-enforced roadmap to compress Gaucher disease therapeutic unit costs by over two orders of magnitude (>= 100x, to JPY 100–1,000 per daily oral dose) while guaranteeing uninterrupted splenic/marrow substrate clearance ("flushing the macrophage lysosomal backlog"). We formalize three synergistic therapeutic architectures: (i) AI-designed non-competitive allosteric pharmacological chaperones targeting the Domain III hydrophobic core of L444P acid beta-glucocerebrosidase (GCase) with binding free energy \Delta G_{bind} <= -4.5 kcal/mol and an acidic lysosomal dissociation switch (pK_a ~ 5.2); (ii) glyco-engineered methylotrophic yeast (Komagataella phaffii) and plant-seed (Oryza sativa) oral micro-encapsulated GCase leveraging M-cell Peyer's patch lymphatic translocation; and (iii) hyper-stabilized de novo engineered Super-GCase (\Delta\Delta G_{fold}^{eng} <= -3.2 kcal/mol) encapsulated in phosphatidylserine/mannose-targeted liposomes. Finally, we integrate this ultra-low-cost therapeutic pipeline with the PHOENIX_ROUTER zero-knowledge hardware interlock over the 64-bit Goldilocks prime field F_p (p = 2^64 - 2^32 + 1) and the NLL-v1.0-2026 / RTO P2.0 optical data diode, physically clamping administrative subsidy-termination write-enable lines (WE_STATE_ROOT = 0V in < 3.2 ns) and preventing unauthorized external AI scraping of Japanese patient haplotype tensors.
