One Oral GLP-1 Receptor Agonist, Three Gateways: What a Nonpeptide Metabolic Candidate's Regulatory Path Reveals About Parallel Health Canada, FDA, and EMA Clinical Trial Authorization

Abstract
Oral, nonpeptide small-molecule glucagon-like peptide-1 (GLP-1) receptor agonists represent a significant pharmaceutical innovation within an already well-established therapeutic mechanism, offering the metabolic benefits long associated with injectable peptide GLP-1 agonists through a once-daily tablet formulation. This article uses a hypothetical multinational Phase 2 clinical trial of an oral GLP-1 receptor agonist candidate, sponsored by a fictional company, to compare the Clinical Trial Application (CTA) process required by Health Canada, the Investigational New Drug (IND) process required by the U.S. Food and Drug Administration, and the Clinical Trial Application process required by the European Medicines Agency under Regulation (EU) No 536/2014. Because this candidate's nonpeptide, small-molecule chemical format places it under each region's conventional small-molecule nonclinical framework rather than the biologics-specific framework governing its peptide-based predecessors, the comparison also addresses a regulatory-classification question of direct practical relevance: how chemical format, not shared pharmacological mechanism, determines which nonclinical and administrative conventions actually apply in each of the three jurisdictions. The article addresses the operational challenges specific to this program, the ways artificial intelligence tools are used, and can responsibly be used, in preparing and administering this kind of regulatory package, and closes with recommendations for regulatory operations teams managing oral metabolic drug candidates across Canada, the United States, and the European Union. Keywords: oral GLP-1 receptor agonist; obesity; type 2 diabetes; Clinical Trial Application; Investigational New Drug; Regulation (EU) No 536/2014; regulatory artificial intelligence
