Abstract
Background
Capivasertib is an oral pan-AKT inhibitor evaluated in combination with multiple systemic therapies across solid tumors, but its efficacy and safety vary by disease setting, biomarker status, and treatment backbone. We aimed to systematically evaluate the efficacy, safety, and early-phase clinical activity of capivasertib-containing combination regimens in advanced breast cancer and other solid tumors.
Methods
MEDLINE, Web of Science, Scopus, Cochrane CENTRAL, trial registries, and major oncology conference sources were searched from inception to 31 May 2026. Randomized comparative studies, single-arm phase II studies, and phase I/Ib combination cohorts were included. Random-effects meta-analyses used restricted maximum likelihood estimation, with randomized and noncomparative evidence synthesized separately.
